Abstract

MEOR is considered more environmentally friendly and economical than other methods because it relies on microbial metabolite products such as biosurfactants, gases, and enzymes. In this study, Bacillus sp. was chosen because of its ability to produce biosurfactants and survive in extreme environmental conditions. To support microbial growth and activity, a carbon source in the form of lactose was added at various concentrations. This study aimed to determine the effect of adding lactose and sucrose additives on the activity of Bacillus sp. in reducing crude oil viscosity for Microbial Enhanced Oil Recovery (MEOR) applications. Bacillus sp. were rejuvenated with various concentrations of lactose (5–20%) and sucrose (5–20%), then incubated at high temperature (60°C) for 3 days to test their survival and potential for biosurfactant production under thermal conditions simulating an oil reservoir environment. The results showed that several treatment combinations, specifically Bacillus sp. A + 20% lactose and Bacillus sp. B + 5% lactose survived incubation at 60°C, as evidenced by the formation of colonies on Nutrient Agar and the Grampositive, rod-shaped morphology observed under a microscope. This viability indicates that both isolates have good thermal tolerance. FTIR analysis revealed the presence of typical biosurfactant functional groups, such as amide I–II, esters, and alkyl chains, indicating bacterial metabolic activity in producing surface-active compounds. This biosurfactant activity correlated with a decrease in oil viscosity, indicating that the bacteria are capable of modifying the oil's physical properties through partial degradation and emulsification. This study concluded that the addition of lactose can increase the resistance and activity of Bacillus sp. at high temperatures and support the production of biosurfactants that are effective in reducing oil viscosity. These Research confirm the potential of Bacillus sp. as MEOR agents in high-temperature reservoirs.
