Abstract

The research titled "Morphological Study of Areca Nut (Areca catechu L.) Varieties Using SCoT Markers in Indragiri Hilir Regency" aims to identify the morphology and analyze the genetic diversity through molecular identification using SCoT markers of five Areca nut varieties found in Indragiri Hilir Regency. The research was conducted in community areca nut plantations in Kuala Lemang village, Keritang district; Kotabaru Seberida village, Keritang district; Mumpa village, Tempuling district; and Pengalihan village, Enok district, Indragiri Hilir Regency, as well as in the Genetics and Molecular Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, University of Riau, Pekanbaru City, for three months from May to July 2024. The research activities included sampling young leaves from five areca nut varieties, morphological observations, total DNA isolation, total DNA electrophoresis, DNA amplification using PCR, PCR product electrophoresis, and data analysis. The results showed that, although there are some morphological differences among the areca nut varieties, the similarities in key traits such as stem height, leaf sheath color, and leaf size suggest that these varieties likely share a common genetic ancestry, with variations arising from environmental adaptation, selective breeding, or natural genetic variation. On a molecular level, the results indicated that although there is considerable genetic variation, the genetic similarities among samples of the same variety are significant enough to form distinct groups in the dendrogram. Additionally, while some samples did not group with the majority of their variety, they still showed significant genetic proximity to the main group.
