Abstract
This study aims to determine the effect of interaction and the main factors of Zeatin and NAA (Naphthalene Acetid Acid) concentration on orchid plant multiplication in vitro. This study was conducted at the Biotechnology Laboratory, Faculty of Agriculture, Riau Islamic University, Jalan Kaharuddin Nasution No.113, Air Dingin Village, Bukit Raya District, Pekanbaru City. The design used in this study was a Factorial Completely Randomized Design consisting of 2 factors and 3 replications. The first factor is the Zeatin factor consisting of 4 treatment levels, namely (0, 3, 6, and 9 ppm) and the second factor is NAA consisting of 4 treatment levels, namely (0, 0.5, 1, and 1.5 ppm). The parameters observed were the percentage of explant life (%), age of shoot emergence (days), number of shoots (fruit), number of roots (fruit), shoot length (cm) and number of somatic embryos (fruit). Observation data from each treatment were analyzed statistically and continued with a further test of Honestly Significant Difference (HSD) at the 5% level. The results showed that the interaction of Zeatin and NAA administration had a significant effect on the age of shoot emergence, number of shoots, number of roots, shoot length and number of somatic embryos. The best treatment was administration of 6 ppm zeatin and 1.5 ppm NAA. The main effect of zeatin had a significant effect on all parameters, the best treatment was administration of 6 ppm zeatin. The main effect of NAA was significant on all parameters, the best treatment was administration of 1.5 ppm NAA.
