Abstract

The purpose of this study was to determine the effect of the interaction between the concentration of zeatin and sucrose growth regulators on the growth of Dendrobium crumenatum swartz callus in vitro. The experimental design used was factorial completely randomized design consisting of 2 factors, namely factor Z (zeatin) with doses (0, 0.1, 1.0, and 10 ppm) and factor S (sucrose) with doses (0, 20, 40, and 60. g / l media), in order to obtain 16 treatment combinations. in each treatment consisted of 3 replications in order to obtain 48 experimental units. Each experimental unit consisted of 4 explants, so that the total explants totaled 192 plants. The observed parameters were as follows: live explant present (%), percentage of rooted explants (%), percentage of explants forming callus (%), number of shoots (fruit), number of leaves (strands), chlorophyll analysis (μg / g). The interaction of zeatin and sucrose administration had an effect on the percentage of live explants, the percentage of explants that formed callus, the number of shoots, and the number of leaves. The best treatment was zeatin 1.0 ppm and sucrose 40 g / l media (Z2S2). The main effect of zeatin has a significant effect on all parameters. The best treatment is zeatin with a dose of 1.0 ppm (Z2). the main effect of sucrose has a significant effect on all parameters. The best treatment is giving sucrose with a dose of 40 g / l medium (S2).
