Abstract

This research focuses on the analysis of several types of matoa (Pometia pinnata) in the educational garden of Siabu village, Salo district, Kampar regency, Riau province, based on morphological characteristics and molecular markers using start codon targeted (SCoT) markers. The study was conducted at the Educational Garden of Siabu Village, Salo District, Kampar Regency, and the Genetics Laboratory of the Biology Department, Faculty of Mathematics and Natural Sciences, University of Riau. The research was carried out over eight months, from July 2023 to February 2024. The research activities included sample collection, morphological observation, total DNA isolation, total DNA electrophoresis, DNA amplification through PCR using SCoT primers, PCR product electrophoresis, and data analysis. The results showed that the matoa plants studied had not produced flowers and fruits, making it impossible to perform a comprehensive morphological analysis. However, the morphology of the roots, stems, and leaves exhibited characteristics similar to those of matoa in general. Additionally, matoa does not yet have a quality standard as stipulated in SNI (Indonesian National Standard) or other applicable quality regulations. At the molecular level, it was found that the green matoa formed a group with the brown-green matoa, while the yellow matoa formed a group with the black and red matoa.
