Abstract
research entitled "The Effect of Concentration of Colchicine on Growth and Production of Black Soybean Varieties Detam 2 (Glycine soja (L) Merr)". The purpose of this study was to determine the concentration of LD50, as well as the effect of colchicine concentration on the growth and yield of black soybean plants of the Detam 2 variety (Glycine soja (L) Merr). The research was conducted at the Cold Water Experimental Garden, Basic Laboratory and Fisheries Laboratory, Faculty of Agriculture, Islamic University of Riau. The time of the research was carried out for six months from January to June 2023. The research consisted of two stages. Phase I research to determine the concentration of colchicine LD50 (Lethal Dose 50%). The treatment concentrations were: K0 = 0 ppm, K1 = 500 ppm, K2 = 1000 ppm, K3 = 1500 ppm, K4 = 2000 ppm, K5 = 2500 ppm, K6 = 3000 ppm, K7 = 3500 ppm. Phase II research was carried out by inducing colchicine mutations using a concentration of 0 (control) and LD47. Phase II research was to determine the effect on growth and production in control plants and plants that had been mutated with LD47 concentrations. Phase I research showed results based on the sensitivity of black soybean plants with a survival percentage of 50%. The concentration of each phase of research was obtained, for the Phase I that is LD50 colchicine was 3599 ppm and the phase II that is LD47 of colchicine was 3500 ppm. The effect of colchicine LD47 significantly affected parameters of petiole length, number of branches, widest leaf area, stem diameter, flowering period, number of pods planted and dry weight of planted seeds. The effect of colchicine LD47 did not significantly affect the parameters of plant height, node or node length, percentage of fruitful pods, length of fruit stalk and weight of 100 dry seeds.
