Abstract

This study aims to identify the effect of colchicine concentration on phenotypic appearance, as well as the effect of colchicine concentration on growth and yield of Rainbow Corn (Zea mays L.) plants. Study carried out at the Cold Water Experimental Garden, Basic Laboratory and Fisheries Laboratory, Faculty of Agriculture, Riau Islamic University. Time The research was carried out for four months from April to July 2023. The research carried out consisted of two stages. Phase one research for determine the colchicine LD50 concentration (Lethal Dose 50%). .The design used in research is. Completely Randomized Design (CRD) with one factor. The treatment concentrations are: without colchicine treatment (C0), 0.01% (C1), 0.05% (C2), 0.1% (C3) and 0.15% (C4). There were 250 experimental seeds in the experiment First. The second phase of research was carried out by inducing colchicine mutations using control and LD48 with a total of 300 seeds. Phase two research carried out to determine the effect on growth and production of control plants and plants that have been mutated with LD48 concentrations. Phase one research shows results based on the sensitivity of corn plants rainbow with seed viability of 50% can be seen that the LD50 concentration is obtained was 0.055% and the concentration used for stage two namely colchicine LD48 concentration of 0.05%. Data from phase 2 research analyzed in the form of quantitative data using the independent sample t-test (T-test). Colchicine LD48 has a significant effect on stem diameter parameters, flowering age, ear weight, ear length, dominant seed color and grade leaf chlorophyll. The effect of colchicine LD48 did not significantly affect the parameters plant height, number of leaves, harvest age and ear diameter.
