Abstract

ABSTRACT Response of Sprouts of Sweet Orange (Citrus sinensis L.) Seeds to In Vitro Addition of BAP and NAA. This research has been carried out at the biotechnology laboratory of the Faculty of Agriculture, Riau Islamic University, Jalan Kaharuddin Nasution Km 11 no. 113, Air Cold Village, Bukit Raya District, Pekanbaru City. The study was conducted from April to July 2020. The purpose of this study was to determine the interaction and main effect of BAP and NAA concentrations on the growth of sweet orange (Citrus sinensis L.) seed sprouts in vitro. This study used a factorial Completely Randomized Design (CRD) consisting of 2 factors, the first factor was BAP consisting of 3 levels, namely 0, 1, and 2 ppm and the second factor was NAA which consisted of 4 levels, namely 0, 1, 2 and 3 ppm. Each treatment combination was repeated 3 times for a total of 36 experimental units. Each experimental unit consisted of 2 culture bottles so that a total of 72 culture bottles were obtained, each bottle consisted of two explants. The average results of each significant parameter were continued with the 5% BNJ test. Parameters observed were growth percentage, age of root emergence, percentage of plantlet formation, shoot height, number of leaves and number of roots. The results showed that the interaction of BAP and NAA had an effect on the parameters of root emergence age, shoot height, number of leaves and number of roots with the best concentration of 1 ppm BAP concentration and 2 ppm NAA concentration. The main effect of BAP concentration was significant on all parameters observed with the best treatment being 1 ppm BAP concentration. The main effect of NAA concentration was real on all observed parameters, the best treatment was 2 ppm NAA concentration.
