Abstract
This research was conducted from September to November 2018. The aim of this study was to determine the outward genetic relationship (Nephelium ramboutanake (Labill.) Leenh) by using five primers based on RAPD (Random Amplified Polymorphic DNA) markers. This research went through several stages, namely sample collection, DNA isolation, quality testing, and data analysis. Data were analyzed using the UPGMA (Unweighted Pair Group Method with Arithmetic Average) method using MVSP 3.22 software. The results of the research on genetic outward relations by using five primers based on RAPD markers resulted in the size of the DNA band length, number of loci, number of bands and dendogram of different outward genotypes. The diversity of the origin of Siak and Bengkalis can be seen from the results of the analysis which has the results of the study obtained the size of the DNA band length which ranges from 150bp - 1500bp. The number of loci is 26 loci, the total number of outward DNA bands is 120 bands. The percentage of similarity (kinship) between the origin of Siak and Bengkalis based on several primers resulted in the level of kinship between the Siak and Bengkalis populations in primary K - 02 80%, primary K - 08 65%, at primary OPJ - 20 65%, primer P - 08 25 %, primer Z - 13 78%. In the population between Siak and Bengkalis produces a kinship relationship of 70 - 100% in the Siak genotype, whereas in the Bengkalis genotype it produces 80 - 100% kinship.
